The determination of various forms of nitrogen compounds in water helps to assess the contamination and "self-purification" of water bodies. Fish are more sensitive to ammonia nitrogen in water.

Ammonia nitrogen (NH3-N) is present in water in the form of free ammonia (NH3) or ammonium salt (NH4+), the composition ratio of which depends on the pH of the water and the temperature of the water. When the pH is high, the proportion of free ammonia is high. On the contrary, the ratio of ammonium salt is high and the water temperature is reversed. The source of ammonia nitrogen in water is mainly the decomposition products of nitrogenous organic matter in domestic sewage by microorganisms, some industrial wastewater, such as coking wastewater and synthetic ammonia fertilizer plant wastewater, and farmland drainage. In addition, in an anaerobic environment, the nitrite present in the water can also be reduced to ammonia by the action of microorganisms. In an aerobic environment, ammonia in water can also be converted to nitrite and even continue to be converted to nitrate.

The ammonia nitrogen present in the form of free ammonia or ammonium ions reacts with the sodium reagent to form a yellow-brown complex, the color of which is proportional to the ammonia nitrogen content. Metal ions such as calcium, magnesium and iron in water, sulfides, aldehydes and ketones, color, and turbidity all interfere with the determination, and corresponding pretreatment is required.

This method can be used for the measurement of water quality such as surface water, industrial wastewater, and domestic sewage.
